I think new innovations within games come along all the time.
Today each new game needs something new to get anything above a 7/10 rating, in fact. Most of theese innovations are pretty small though: the movement system of Assassin's Creed, the dismemberment system of Dead Space and so on.
There have been a few major innovations lately though: the third person action games. Gears of War popularised the genre extremely and now I think most action games are third person actually. Gears also introducted the cover system that now is standard for action games (and is beeing experimented with for FPS games in Killzone 2) and popularised two player games. There are more two player games now than in a long time: Gears of War, Army of Two, Kane and Lynch, Fable II - the two first working a bit better, the two latter a bit worse.
Mirror's Edge did something completely fresh aswell, but sadly I don't think it will be enough to start a new genre. It will probably go the way of Hack n' Slash games (The Devil May Cry and Ninja Gaiden series) and never become largely popular as Mirror's Edge alone pretty much fills all our third person acrobatics needs.
I have to mention Portal aswell. That one did something completely new and did it damn well.
